The Biacore 3000 has all advantages of a Biacore 2000 instrument with the benefit of higher sensitivity, shorter analysis time and lower sample consumption.
Compared to a Biacore 2000 the detection unit has a reduced signal to noise ratio which allows an easier detection of peptide and small molecule interactions with lower molecular weight. Furthermore, the dynamic range of the detection unit is enhanced enabling interaction analyses in the presence of buffer additives with high refractive index. The microfluidic system consists of four serially directed flow cells, but in contrast to the Biacore 2000 the height of the flow cells is reduced. Thus, faster kinetics can be measured with less interference from mass transfer limitations (kass from 103 - 107 M-1 s-1 and kdiss from 10-1 - 10-6 s-1). The molecular detection limit for small molecules is about 180 Da with an analytical performance of 1 pM.
The sample recovery function from bound material is enhanced, facilitating mass spectrometry analyses.
The Biacore 3000 system is ideally suited for proteins, peptides and even small molecules with weak or fast kinetics. Biaffin uses one Biacore 3000 to study more evaluated binding kinetics.
